Understanding Melanocortin Receptors: MC1R to MC5R
The melanocortin receptor family consists of five G-protein-coupled receptors — MC1R through MC5R — each with a distinct tissue distribution and physiological role. Understanding which receptors a given peptide engages is central to interpreting melanocortin research, since compounds with different receptor profiles can produce very different downstream effects.
The Five Receptors at a Glance
- MC1R: primarily expressed in melanocytes; the main driver of pigmentation-related signalling.
- MC2R: found in the adrenal cortex; mediates ACTH signalling rather than α-MSH.
- MC3R: expressed in the brain and periphery; implicated in energy homeostasis and inflammatory regulation.
- MC4R: primarily central nervous system; extensively studied in appetite and energy-balance research.
- MC5R: broadly expressed peripherally, including exocrine tissue; comparatively less well characterised than the others.
Why Receptor Selectivity Matters in Research
A compound's receptor selectivity profile determines which experimental readouts are relevant. Melanotan 1, being largely MC1R-selective, is typically used where pigmentation-pathway specificity matters. Melanotan 2's broader MC1R/MC3R/MC4R/MC5R profile makes it more suited to studies spanning multiple melanocortin pathways.
